The effects of medium and sterilization on the mature embryo culture of barley (Hordeum vulgare L.) were studied. Among the sterilization methods studied, sodium hypochlorite sterilization followed by treatment of antibiotic solution and sterilization HgCl2 application was found to be more effective than the other methods. Arpa (Hordeum vulgare L.)’da Embriyo Kültürü Özet: Arpa (Hordeum vulgare L.)’n›n olgun embriyolar›n›n kültüründe besin ortam› bileflimi ve sterilizasyon yönteminin etkisi üzerinde durulmufltur. Kullan›lan besin ortamlar› içerisinde embriyodan bitkicik geliflimi en yüksek Randolph-Cox ortam›nda elde edilmifltir (%92,1). Bunu s›ras›yla Murashige-Skoog (MS) (%91,3); 1/2 MS (%87,7) ve B5 (%85,7) ortamlar› izlemifltir. Araflt›r›lan sterilizasyon yöntemlerinden sodyum hipoklorit ile sterilizasyonu antibiyotik solusyonu ile muamele etme ve HgCl2 ile yap›lan sterilizasyon daha etkili bulunmufltur. Ancak, HgCl2’ün embriyodan bitkicik geliflimi üzerine istatistiki bak›mdan önemli olmayan olumsuz etkisi de gözlenmifltir.
