TAILIEUCHUNG - Đa dạng di truyền loài giổi ăn hạt (Michelia tonkinensis) tại khu rừng thực nghiệm, trường Đại học Lâm nghiệp dựa trên chỉ thị phân tử RAPD

Với 22 mẫu giổi ăn hạt (Michelia tonkinensis A. Chev.) | TAP CHI SINH HOC 2019 41 2se1 amp 2se2 419 426 DOI 0866-7160 v41n2se1 amp GENETIC DIVERSITY OF Michelia tonkinensis . Magnoliaceae AT THE EXPERIMENTAL FOREST VIETNAM NATIONAL UNIVERSITY OF FORESTRY BASED ON RAPD MARKERS Vu Quang Nam Nguyen Thi Tho Nguyen Thi Hai Ha Tran Van Dang College of Forestry Biotechnology Vietnam National University of Forestry Ha Noi Vietnam Received 12 August 2019 accepted 29 September 2019 ABSTRACT Twenty two samples of Michelia tonkinensis at 5 8 18 years old originated from Ha Noi Ba Vi Hoa Binh and Ninh Bình provinces and were introduced into Experimental Forest Vietnam National University of Forestry VNUF were used to study genetic diversity based on 09 RAPD primers OPAC14 OPF9 OPA7 OPA20 OPB10 RA142 RA159 OPD11 and RA46 . The results showed that the total 1138 bands generated 786 of which were found to be polymorphic. The number of amplification products ranged from 5 to 11 for different primers. Polymorphic Information Content PIC of these primers are low from OPA7 primer to OPAC14 primer with average of . Genetic similarity coefficients are ranged from to with average of . This shows the degree of genetic similarity of the Michelia tonkinensis samples in the experimental forest of VNUF is high. Thus it is created to be heterotic in sexual reproduction but it is difficult to make the genetic disversity in the group of Michelia tonkinensis in VNUF. UPGMA cluster analysis place these samples into two main groups of which group I had only G6 sample group II consisted of remain 21 samples and further devided into two subgroups with subgroup 1 including G1 and G16 and subgroup 2 with the remaining 19 samples and further devided into 2 clusters cluster 1 G2 G3 G4 G5 G19 and cluster 2 G8 G10 G13 G14 G7 G9 G11 G12 G15 G17 G18 G20 G21 G22 . The study reveals the limitation of genetic basis of the current Michelia tonkinensis in VNUF and the need to genetically diversity by using new
