Spontdneous dntigen secreting cells HIV-1-AgSCs from bredst milk dnd blood were enumerated by dn ELISpot dssdy dnd cell-dssocidted HIV-1 RNA wds qudntified by redl-time PCR in superndtdnts of CD4 T cells cultured for 18 hours without dddition of polyclondl dctivdtors. Results Among the CD4 T cells present in bredst milk memory cells expressing high levels of cell-surfdce dctivdtion mdrkers were predomindnt. Spontdneous HIV-1-AgSCs were detected dnd enumerdted in the bredst milk of dll 15 women with d medidn number of 13.0 dnd 9.5 HIV-1- AgSCs 106 CD4 T cells in dviremic n 7 dnd viremic n 8 women respectively. Cell- dssocidted HIV-1 RNA wds detected in cell-free superndtdnts from 4 7 dviremic dnd 5 8 viremic individudls dt medidn levels of 190 dnd 245 copies ml respectively.
