In this study we investigated the impact of post-traumatic hypoxia in ampiifying secondary brain damage using a rat modei of diffuse traumatic axonai injury TAI . Rats were examined for behaviorai and sensorimotor deficits increased brain production of inflammatory cytokines formation of cerebrai edema changes in brain metaboiism and eniargement of the iaterai ventricies. Methods Aduit maie Sprague-Dawiey rats were subjected to diffuse TAI using the Marmarou impact-acceieration modei. Subsequentiy rats underwent a 30-minute period of hypoxic 12 O2 88 N2 or normoxic 22 O2 78 N2 ventiiation. Hypoxia-oniy and sham surgery groups without TAI received 30 minutes of hypoxic or normoxic ventiiation respectiveiy. The parameters examined inciuded 1 behaviourai and sensorimotor deficit using the Rotarod beam waik and adhesive tape removai tests and voiuntary open fieid expioration behavior 2 formation of cerebrai edema by the wet-dry tissue weight ratio method 3 eniargement of the iaterai ventricies 4 production of inflammatory cytokines and 5 reai-time brain metaboiite changes as assessed by microdiaiysis technique. Results TAI rats showed significant deficits in sensorimotor function and deveioped substantiai edema and ventricuiar eniargement when compared to shams.
